Intended use
     This immunoassay kit allows for the specific measurement of bovine Leptin concentrations in cell culture supernates, serum, and plasma.
 
Introduction
     Leptin (from the Greek leptos, meaning thin) is a protein hormone with important effects in regulating body weight, metabolism and reproductive function. The protein is approximately ~16 kDa in mass and encoded by the obese (ob) gene. Leptin is expressed predominantly by adipocytes, which fits with the idea that body weight is sensed as the total mass of fat in the body. Smaller amounts of leptin are also secreted by cells in the epithelium of the stomach and in the placenta. Leptin receptors are highly expressed in areas of the hypothalamus known to be important in regulating body weight, as well as in T lymphocytes and vascular endothelial cells. 
Test principle
     This assay employs the quantitative sandwich enzyme immunoassay technique. A monoclonal
antibody specific for Leptin has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any Leptin present is bound by the immobilized antibody. An
enzyme-linked monoclonal antibody specific for Leptin is added to the wells. Following a wash to remove any unbound antibody-enzyme reagent, a substrate solution is added to the wells
and color develops in proportion to the amount of Leptin bound in the initial step. The color
development is stopped and the intensity of the color is measured.
