ELISA法定量测定大鼠血清、血浆、或其它相关液体中雌激素含量。
概述
雌激素（estrogen）是一类由内分泌产生的类固醇，主要促进女性生殖器官发育和第二性征的出现。其主要由卵巢分泌，可分为雌酮（E1）、雌二醇（E2）和雌三醇（E3）。其中E2的生物活性最强。雌激素扩散进入靶细胞与雌激素受体（ER）结合，影响靶组织的生长期、分化与功能。
实验原理
本试剂盒应用竞争抑制酶标免疫分析法测定标本中雌激素水平。用纯化的抗体包被微孔板，制成固相抗体，固相抗体与一定量的生物素标记的雌激素和非标记抗原(标本或标准品)进行竞争抑制反应，待测定的标本量越多，抗体与生物素标记的雌激素结合就越少，反之结合就多。最后再加入HRP标记的亲和素，经过彻底洗涤后用底物TMB显色。TMB在过氧化物酶的催化下转化成蓝色，并在酸的作用下转化成最终的黄色。颜色的深浅和样品中的雌激素呈负相关。用酶标仪在450nm波长下测定吸光度（OD值），计算样品浓度。
Intended use
This immunoassay kit allows for the specific measurement of total estrogen concentrations of natural and recombinant rat estrogen in plasma and serum.
Introduction
Natural estrogens are hormones secreted principally by the ovarian follicles and also by the adrenals, corpus luteum, placenta . Exogenous estrogens (natural or synthetic) elicit, to varying degrees, all the pharmacologic responses usually produced by endogenous estrogens. Estrogenic hormones are secreted at varying rates during the menstrual cycle throughout the period of ovarian activity. The gonadotropins of the anterior pituitary regulate secretion of the ovarian hormones, estradiol and estrogen; hypothalamic control of pituitary gonadotropin production is in turn regulated by plasma concentrations of the estrogens and estrogen. During pregnancy, the placenta becomes the main source of estrogens. At the menopause, ovarian secretion of estrogens declines at varying rates.
Test principle
This assay employs the competitive inhibition enzyme immunoassay technique. A monoclonal antibody specific for estrogen has been pre-coated onto a microplate. A competitive inhibition reaction is launched between biotin labeled estrogen and biotin unlabeled antigens (Standards or samples) with the pre-coated antibody specific for estrogen. The more the amount of ESTROGEN in samples, the less the biotin labeled estrogen bound by pre-coated antibody. Vice versa. Then HRP labeld avidin and substrate solution are added to the wells, respectively. And the color develops in opposite to the amount of ESTROGEN bound in the initial step. The color development is stopped and the intensity of the color is measured. |
