ELISA法定量测定人血清、血浆或其它相关液体中细胞表面膜联蛋白V (annexin V)含量。
 
实验原理
Annexin V是一种分子量为35~36kD的Ca2+依赖性磷脂结合蛋白,与磷脂酰丝氨酸有高度亲和力,故可通过细胞外侧暴露的磷脂酰丝氨酸与凋亡早期细胞的胞膜结合。因此Annexin V被作为检测细胞早期凋亡的灵敏指标之一。
本试剂盒应用双抗体夹心酶标免疫分析法测定标本中Annexin V水平。用纯化的抗体包被微孔板,制成固相抗体,往包被单抗的微孔中依次加入Annexin V抗原、生物素化的抗人Annexin V抗体、HRP标记的亲和素,经过彻底洗涤后用底物TMB显色。TMB在过氧化物酶的催化下转化成蓝色,并在酸的作用下转化成最终的黄色。颜色的深浅和样品中的Annexin V呈正相关。用酶标仪在450nm波长下测定吸光度(OD值),计算样品浓度。
Intended use
This immunoassay kit allows for the specific measurement of Human Annexin V concentrations in cell culture supernates, serum and plasma.
 
Introduction
Annexin V is a calcium binding protein which is widely present in various cells and tissues. Annexin V, belonging to a recently discovered family of proteins, the annexins, with anticoagulant properties has proven to be a useful tool in detecting apoptotic cells since it preferentially binds to negatively charged phospholipids like PS in the presence of Ca2+ and shows minimal binding to phosphatidylcholine and sphingomyeline. Changes in phosphatidylserine(PS) asymmetry, which is analyzed by measuring Annexin V binding to the cell membrane, were detected before morphological changes associated with apoptosis have occurred and before membrane integrity has been lost. By conjugating FITC to Annexin V it is possible to identify and quantitate apoptotic cells on a single-cell basis by flow cytometry. Staining cells simultaneously with FITC-Annexin V (green fluorescence) and the non-vital dye propidium iodide (red fluorescence) allows (bivariate analysis) the discrimination of intact cells (FITC-PI-), early apoptotic (FITC+PI-) and late apoptotic or necrotic cells (FITC+PI+).
 
Test principle
This assay employs the quantitative sandwich enzyme immunoassay technique. A monoclonal antibody specific for Annexin V has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any Annexin V present is bound by the immobilized antibody. An enzyme-linked monoclonal antibody specific for Annexin V is added to the wells. Following a wash to remove any unbound antibody-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of Annexin V bound in the initial step. The color development is stopped and the intensity of the color is measured.
