Introduction Japanese encephalitis virus (JEV) is a mosquito-borne Flavivirus that is closely related to West Nile and St. Louis encephalitis viruses. JEV infection occurs throughout most of Asia and is the most common cause of encephalitis in that region. Other than sporadic travel-associated cases, Japanese encephalitis (JE) has not been identified in Africa, Europe or the Americas. Test principle This assay employs the quantitative sandwich enzyme immunoassay technique. Inactivated JE antigen has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any JE IgG present is bound by the immobilized antibody. An enzyme-linked monoclonal antibody specific for JE IgG is added to the wells. Following a wash to remove any unbound antibody-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of JE IgG bound in the initial step. The color development is stopped and the intensity of the color is measured. |
