| 预期应用 ELISA法定量测定犬血清、血浆或其它相关液体中LH含量。   实验原理 本试剂盒应用竞争抑制酶标免疫分析法测定标本中LH水平。用纯化的抗体包被微孔板,制成固相抗体,固相抗体与一定量的生物素标记的LH和非标记抗原(标本或标准品)进行竞争抑制反应,待测定的标本量越多,抗体与生物素标记的LH结合就越少,反之结合就多。最后再加入HRP标记的亲和素,经过彻底洗涤后用底物TMB显色。TMB在过氧化物酶的催化下转化成蓝色,并在酸的作用下转化成最终的黄色。颜色的深浅和样品中的LH呈负相关。用酶标仪在450nm波长下测定吸光度(OD值),计算样品浓度。    Intended use This immunoassay kit allows for the specific measurement of total concentrations of natural and recombinant canine LH in plasma and serum.   Introduction Luteinizing hormone (LH) is an important hormone of the reproductive system, which has found application in diagnosis and therapeutic medicine. It plays a vital role in the development and functioning of the reproductive system. Determination of LH concentration is important for detection of dysfunction of the pituitary variance axis, diagnosis of reproductive disorders, monitoring of antifertility programmes and in therapeutic preparations.   Test principle This assay employs the competitive inhibition enzyme immunoassay technique. A monoclonal antibody specific for LH has been pre-coated onto a microplate. A competitive inhibition reaction is launched between biotin labeled LH and biotin unlabeled antigens (Standards or samples) with the pre-coated antibody specific for LH. The more the amount of LH in samples, the less the biotin labeled LH bound by pre-coated antibody. Vice versa. Then HRP labeld avidin and substrate solution are added to the wells, respectively. And the color develops in opposite to the amount of LH bound in the initial step. The color development is stopped and the intensity of the color is measured.  |
