| 预期应用     ELISA法定量测定兔血清、血浆、细胞培养物上清或其它相关液体中NE含量。   概述     肾上腺素(adrenaline,epinephrine,AD)是肾上腺髓质的主要激素,其生物合成主要是在髓质铬细胞中首先形成去甲肾上腺素,然后进一步经苯乙胺-N-甲基转移酶(phenylethanolamine N-methyl transferase,PNMT)的作用,使去甲肾上腺素甲基化形成肾上腺素。正常肾上腺髓质的分泌物中,去甲肾上腺素仅占10%~18%,但尿中和血浆中去甲肾上腺素约为肾上腺素的5倍。而嗜铬细胞瘤分泌物中去甲肾上腺素可达50%~90%,说明嗜铬细胞瘤分泌的儿茶酚胺以去甲肾上腺素为主。   实验原理     本试剂盒应用双抗体夹心酶标免疫分析法测定标本中NE水平。用纯化的抗体包被微孔板,制成固相抗体,往包被单抗的微孔中依次加入NE、生物素化的抗人NE抗体、HRP标记的亲和素,经过彻底洗涤后用底物TMB显色。TMB在过氧化物酶的催化下转化成蓝色,并在酸的作用下转化成最终的黄色。颜色的深浅和样品中的NE呈正相关。用酶标仪在450nm波长下测定吸光度(OD值),计算样品浓度。    Intended use     This immunoassay kit allows for the specific measurement of Rabbit Noradrenalin concentrations in cell culture supernates, serum, and plasma.   Introduction     The catecholamines adrenalin, noradrenalin and dopamine are synthesized in the adrenal medulla, the sympathetic nervous system and in the brain. They influence virtually all tissues and are involved together with other hormonal and neuronal systems in the regulation of a wide variety of physiological processes. As catecholamines and their metabolites metanephrine and normetanephrine are secreted in increasing amounts in a number of diseases, they may be used for diagnostic purposes. In this context, diagnosis as well as the follow-up of tumor diseases of the nervous system are of special importance. This applies primarily to the pheochromocytoma, but also the neuroblastoma and the ganglioneuroma.   Test principle     This assay employs the quantitative sandwich enzyme immunoassay technique. A monoclonal antibody specific for Noradrenalin has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any Noradrenalin present is bound by the immobilized antibody. An enzyme-linked monoclonal antibody specific for Noradrenalin is added to the wells. Following a wash to remove any unbound antibody-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of Noradrenalin bound in the initial step. The color development is stopped and the intensity of the color is measured.
