| 预期应用     ELISA法定量测定人血清、血浆、细胞培养物上清或其它相关液体中sPECAM-1含量。   实验原理     本试剂盒应用双抗体夹心酶标免疫分析法测定标本中sPECAM-1水平。用纯化的抗体包被微孔板,制成固相抗体,往包被单抗的微孔中依次加入sPECAM-1抗原、生物素化的抗人sPECAM-1抗体、HRP标记的亲和素,经过彻底洗涤后用底物TMB显色。TMB在过氧化物酶的催化下转化成蓝色,并在酸的作用下转化成最终的黄色。颜色的深浅和样品中的sPECAM-1呈正相关。用酶标仪在450nm波长下测定吸光度(OD值),计算样品浓度。   Intended use     This immunoassay kit allows for the specific measurement of human sPECAM-1 concentrations in cell culture supernates, serum, and plasma.   Introduction     PECAM-1 (platelet endothelial cell adhesion molecule-1) also called CD31 and EndoCAM belongs to the immunoglobulin superfamily. PECAM-1 is a transmembrane glycoprotein with a molecular weight of approximately 130 kDa, depending on the degree of glycosylation . Bone marrow stem cells and transformed cell lines of the myeloid and megakaryocytic lineage express PECAM-1. PECAM-1 was also detected in human, mouse and rat solid tumor lines. Studies have suggested a role for PECAM-1 in the inflammatory process and leukocyte-endothelial interaction. The process of leukocyte emigration to the site of inflammation can be dissected into three  successive stages: (1) rolling, mediated by the selectins; (2) tight adhesion mediated by ICAMs and their counter-receptors, the integrins; and (3) transmigration of leukocytes through  intercellular junctions of vascular endothelial cells which requires PECAM-1. PECAM-1 is an early and sensitive marker for tumor-induced angiogenesis. Several data have suggested that PECAM-1 may be involved in the process of angiogenesis in a developing vertebrate embryo as well as during metastases formation. Soluble PECAM-1, which is detected in normal human plasma, is encoded by an alternatively spliced mRNA, from which the exon containing the transmembrane domain has been removed. The decrease levels of sPECAM-1 are detected in patients with cancer.   Test principle     This assay employs the quantitative sandwich enzyme immunoassay technique. A monoclonal antibody specific for sPECAM-1has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any sPECAM-1present is bound by the immobilized antibody. An enzyme-linked monoclonal antibody specific for sPECAM-1is added to the wells. Following a wash to remove any unbound antibody-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of sPECAM-1bound in the initial step. The color development is stopped and the intensity of the color is measured.
