| 预期应用 ELISA法定量测定人血清、血浆、细胞培养物上清或其它相关液体中人5-羟色氨(5-HT) 含量。 实验原理 5-羟色胺(5-hydroxytryptamine,5-HT),又名血清素(serotonin),作为自体活性物质,约90%合成和分布于肠嗜铬细胞,通常与ATP等物质一起储存于细胞颗粒内。在刺激因素作用下,5-HT从颗粒内释放、弥散到血液,并被血小板摄取和储存,储存量约占全身的8%。5-HT作为神经递质,主要分布于松果体和下丘脑,可能参与痛觉、睡眠和体温等生理功能的调节。中枢神经系统5-HT含量及功能异常可能与精神病和偏头痛等多种疾病的发病有关。 本试剂盒应用双抗体夹心酶标免疫分析法测定标本中5-HT水平。用纯化的抗体包被微孔板,制成固相抗体,往包被单抗的微孔中依次加入5-HT抗原、生物素化的抗人5-HT抗体、HRP标记的亲和素,经过彻底洗涤后用底物TMB显色。TMB在过氧化物酶的催化下转化成蓝色,并在酸的作用下转化成最终的黄色。颜色的深浅和样品中的5-HT呈正相关。用酶标仪在450nm波长下测定吸光度(OD值),计算样品浓度。  Intended use This immunoassay kit allows for the specific measurement of human Serotonin 5-Hydroxytryptamine(5-HT) concentrations in cell culture supernates, serum, and plasma. For example, medications that affect the action of serotonin are used to treat depression  Test principle This assay employs the quantitative sandwich enzyme immunoassay technique. A monoclonal antibody specific for 5-HT has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any 5-HT present is bound by the immobilized antibody. An enzyme-linked monoclonal antibody specific for 5-HT is added to the wells. Following a wash to remove any unbound antibody-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of 5-HT bound in the initial step. The color development is stopped and the intensity of the color is measured.
