- 脑钠素(BNP)是继心钠素(ANP)后发现的另一种心脏利钠肽，主要是由心室分泌，与ANP具有相似的扩张血管、利尿、利钠、抑制醛固酮分泌及交感神经活性等广泛的心血管作用，能较敏感地反映心功能状态。随着定量脑钠素测定诊断心力衰竭方案的不断完善，该研究工作为诊断心力衰竭以及鉴别肺源性呼吸困难和心源性呼吸困难提供了有效的方法。

- 实验原理
- 本试剂盒应用双抗体夹心酶标免疫分析法测定标本中BNP水平。用纯化的抗体包被微孔板，制成固相抗体，往包被单抗的微孔中依次加入BNP抗原、生物素化的抗人BNP抗体、HRP标记的亲和素，经过彻底洗涤后用底物TMB显色。TMB在过氧化物酶的催化下转化成蓝色，并在酸的作用下转化成最终的黄色。颜色的深浅和样品中的BNP呈正相关。用酶标仪在450nm波长下测定吸光度（OD值），计算样品浓度。

- Introduction
- Brain natriuretic peptide (BNP, also known as B-type natriuretic peptide or "GC-B") is a natriuretic hormone that is similar to ANP. It is a 32-amino-acid polypeptide secreted by the ventricles of the heart in response to excessive stretching of myocytes (heart muscles cells). BNP is a valuable marker in heart failure and its therapy, for example cardiac resynchronization therapy.

- Test principle
- This assay employs the quantitative sandwich enzyme immunoassay technique. A monoclonal antibody specific for BNP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any BNP present is bound by the immobilized antibody. An enzyme-linked monoclonal antibody specific for BNP is added to the wells. Following a wash to remove any unbound antibody-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of BNP bound in the initial step. The color development is stopped and the intensity of the color is measured.
